Abstract

In this study, molecular species identification of Potato Cyst Nematode (PKN) populations taken from 38 locations where intensive potato cultivation is carried out in Niğde province was made. Golden cyst nematode named Globodera rostochiensis (Wollenweber, 1923) Skarbilovich, 1959 and White cyst nematode named G. pallida Stone, 1973 (Tylenchida: Heteroderidae) are species that cause serious yield losses in potato cultivation in our country and worldwide. G. rostochiensis, which was first detected in Bolu province in Turkey in 1985, was later detected in Niğde, Nevşehir, İzmir and Afyon provinces in the following years. Especially G. rostochiensis and G. pallida are international quarantine pests and their accurate and rapid diagnosis is very important in developing resistant varieties and determining control strategies. ITS5, PITSp4 and PITSr3 primers were used to distinguish nematode species by molecular method. As a result of species identification, G. pallida was not detected and all samples were identified as G. rostochiensis.
